Profinity IMAC 介質(zhì)是一種用于純化帶有多 His 標(biāo)簽的重組蛋白 ( His–tagged proteins ) 的親和介質(zhì)。IMAC 是一種非常有效的分離技術(shù)，它利用組氨酸能夠與金屬離子發(fā)生親和作用的原理對(duì)蛋白質(zhì)加以分離。Profinity IMAC 介質(zhì)以UNOsphere™ 為基質(zhì)，含有作為二價(jià)、三價(jià)金屬離子螯合配位體的亞氨基二乙酸 ( IDA )。當(dāng)配位體結(jié)合有 Zn2+、Ni2+或 Cu2+等金屬離子時(shí)，含組氨酸的蛋白會(huì)與此層析介質(zhì)結(jié)合。
Profinity IMAC 在高流速下具有很高的載量，能在變性和非變性條件下使用，適合于在液相層析系統(tǒng)，重力柱和微型離心柱上使用。
Bio-Rad 提供兩種類型的介質(zhì)：螯合 Ni2+和未螯合金屬離子。未螯合金屬離子的介質(zhì)可以根據(jù)需要螯合任何金屬離子，以zui大化實(shí)現(xiàn)純化的靈活性。
